目的 :探讨中药方剂参附汤对HL6 0细胞热休克反应时热休克蛋白信使核糖核酸 (heatshockproteinmRNA ,HSPmRNA)和糖皮质激素受体信使核糖核酸 (glucocorticoidreceptormRNA ,GRmRNA)表达及对细胞存活率的影响。方法 :建立细胞热休克模型 ,用动物血清学方法制备参附汤 (SFT)血清 ,用Northernblot分析HSPmRNA和GRmRNA的改变 ,用台盼蓝拒染法观察细胞存活率。结果 :HL6 0细胞热休克反应时GRmRNA明显减少 ,细胞存活率下降 ,HSPmRNA则增加 ;该细胞经SFT处理后在热休克反应时 ,GRmRNA表达增加 ,细胞存活率提高 ,HSPmRNA进一步增加。结论 :SFT可上调HL6 0细胞热休克反应时HSPmRNA和GRmRNA表达 ,提高糖皮质激素的生物学效应和抗细胞损伤能力 ,从而提高HL6 0细胞在热休克时的存活率 ,这可能是临床上SFT用于治疗失血性休克的机制之一。
Objective: To investigate the effects of Chinese herbal prescription Shenfu decoction on the expression of heat shock protein mRNA (HSPmRNA) and glucocorticoid receptor mRNA (GRmRNA) in heat shock response of HL60 cells and their effects on cell survival. METHODS: A cell heat shock model was established. SFT serum was prepared by animal serology. Changes of HSP mRNA and GR mRNA were analyzed by Northern blot. The survival rate of cells was observed by trypan blue exclusion. RESULTS: GR mRNA was significantly decreased, cell survival rate was decreased, and HSP mRNA was increased in heat shock response of HL60 cells. In response to heat shock, the expression of GR mRNA was increased, cell survival rate was increased, and HSP mRNA was further increased. Conclusion: SFT can up-regulate the expression of HSP mRNA and GR mRNA in heat shock response of HL60 cells, increase the biological effect of glucocorticoid and anti-cell injury, and increase the survival rate of HL60 cells in heat shock. This may be a clinical SFT. One of the mechanisms used to treat hemorrhagic shock.
