目的 研究输血传播肝炎病毒 (TTV)有否直接垂直传播途径及尿液传播的可能性 ,初步了解西安地区 TTV结构部分特点 .方法 参照文献 [1]在 TTV DNA ORF1区设计一套套式 PCR引物 ,建立 TTV DNA套式 PCR检测方法 ,对 16例血清检测TTV DNA阳性的患者尿液做了 TTV DNA检测 ,对 5 2例正常顺产孕妇血清及其婴儿脐带血同时检测 TTV DNA;并且对西安地区 1例原因不明而 TTV阳性肝炎患者的 TTV套式 PCR扩增终产物纯化直接测序 .结果 16例血清检测 TTV DNA阳性的患者尿液 TTV DNA检测未发现 1例阳性 ;在 5 2例顺产妇外周血中 ,9例 TTV DNA阳性 TTV基因检测阳性率为 17.3%(9/5 2 ) ;而新生儿脐带血 TTV基因检测阳性率为 7.7%(4 / 5 2 ) ;其中 ,母子同时 TTV DNA阳性 2例 ,单纯母亲血 TTV DNA阳性 7例 ;单纯婴儿脐带血 TTV DNA阳性 2例 .直接测序法测序 ,测得的 143bp序列用 DNAsis分析软件中国 1株 (BDH1)序列比较 ,二者同源性为 6 5 .0 %,差异率为 35 .0 %(>30 %) .结论 TTV可能不能通过尿液传播 ,但能经胎盘直接垂直传播 ,西安地区TTV流行毒株可能存在新的 TTV基因型 .
Objective To study whether there is a direct transmission of hepatitis T virus (TTV) and the possibility of urine transmission, so as to understand the characteristics of TTV in Xi’an area.Methods A set of primers was designed based on the ORF1 region of TTV DNA, A TTV DNA nested PCR method was established to detect TTV DNA in urine from 16 TTV DNA positive patients and TTV DNA from 52 normal pregnant women and their umbilical cord blood samples. Cases of unknown cause and TTV positive hepatitis patients by TTV nested PCR amplification of the final product of direct sequencing.Results 16 cases of serum TTV DNA positive urine TTV DNA test did not find a positive case; 52 cases of mothers peripheral blood , The positive rate of TTV DNA positive TTV gene was 17.3% (9/5 2) in 9 cases, while the positive rate of TTV gene in neonatal cord blood was 7.7% (4/52). There may be new TTV genotype.
