目的从脑卒中后大鼠精神症状组群的改变探讨脑卒中后精神症状的演变趋势及益肾调气方药——颐脑解郁方对其早期干预作用。方法选择雄性Wistar大鼠,经Open-Field行为学评分后,随机分为正常组、假手术组、多发脑梗(MCI)组、中药组、西药组、卒中后抑郁(PSD)组及血管性痴呆(VD)组。正常组常规饲养。假手术组经颈外动脉推注0.3 mL生理盐水,并灌胃蒸馏水。其余各组均采用同种系微栓子体外注入法,由颈外动脉推注同等剂量的同种异体血栓栓子;中药组及西药组在模型制备后,予颐脑解郁方、尼莫地平干预给药4周;PSD组术后7 d予21 d慢性不可预知的温和应激(CUMS)加孤养复制PSD模型;VD组术后28 d经水迷宫筛选后进入实验。模型复制成功后,于第4周进行表征观察包括动态观察大鼠毛发、眼部变化,平均进食量及大鼠体重;同时进行行为学测试:蔗糖水消耗实验、Open-Field行为测定、穿梭箱主动回避反应实验。结果表征观察发现3个模型组大鼠平均进食量下降,尤以PSD组及VD组下降显著,与体重变化趋势相一致。行为学测定发现3个模型组大鼠存在兴趣及记忆能力明显下降;蔗糖水消耗量、Open-Field实验提示PSD组以兴趣下降为主;穿梭箱主动回避反应实验结果提示VD组记忆及学习能力下降更为明显。而中药能减缓表征、学习和兴趣能力的下降趋势。结论脑血管病后大鼠存在精神症状组群的改变,并有向PSD及VD演变趋势。颐脑解郁方能减慢此过程的进展。
OBJECTIVE: To investigate the evolution trend of mental symptoms after stroke and the effect of Yinao Tiaoyu Recipe (YNXTC) on its early intervention. Methods Male Wistar rats were randomly divided into normal group, sham operation group, MCI group, TCM group, western medicine group, post-stroke depression (PSD) group and vascularity Dementia (VD) group. Normal group of conventional breeding. In the sham-operated group, 0.3 mL of normal saline was injected through the external carotid artery and distilled water was infused. All the other groups were injected with the same kind of micro-emboli by in vitro injection of the same dose of allogeneic thromboembolism from the external carotid artery. After the preparation of the model group, the rats were treated with Yinao Jieyu Decoction and nimodipine The rats in PSD group were treated with CUMS (unpredictable mild stress) for 21 days, and isolated and isolated PSD models on the 7th day after surgery. Results The results showed that the average food intake of the three model groups decreased significantly, especially in the PSD group and VD group decreased significantly, consistent with the body weight trend. Behavioral testing found that rats in the three model groups were significantly decreased in their interest and memory capacity; sucrose water consumption, Open-Field experiments suggested that interest in PSD group decreased mainly; shuttle box active avoidance reaction showed VD group memory and learning ability Decline is even more obvious. While traditional Chinese medicine can slow down the downward trend of characterization, learning and interest ability. Conclusions There is a change of psychotic group in rats after cerebrovascular disease, and there is a trend toward evolution of PSD and VD. Yinao Jieyu can slow the progress of this process.
