目的观察羟基喜树碱对人骨髓增生异常综合征(MDS)MUTZ-1细胞体外生长的影响。方法采用四甲基偶氮唑蓝(MTT)比色法,测定羟基喜树碱对人骨髓增生异常综合征MUTZ-1细胞生长的抑制作用,并绘制浓度抑制率曲线;采用流式细胞技术检测药物诱导的凋亡细胞量;光镜及透射电镜观察羟基喜树碱处理后细胞形态学的变化。结果羟基喜树碱对MUTZ-1细胞生长抑制作用呈现剂量和时间依赖性,其作用不同时间的半数抑制浓度(IC50)分别为91.198、8.983、6.204、2.509、0.364。羟基喜树碱可诱导MUTZ-1细胞凋亡,呈现剂量依赖性。光镜下可见凋亡细胞胞膜完整、胞体固缩、核固缩、核染色质碎裂;透射电镜下可见凋亡细胞核染色质凝聚、边集,部分细胞核膜消失,细胞浆浓缩、密度增加,浆内可见大小不规则的染色质团块。结论羟基喜树碱对MUTZ-1细胞生长有明显的抑制作用,并可诱导MUTZ-1细胞凋亡。
Objective To observe the effect of hydroxycamptothecin on the growth of human myelodysplastic syndrome (MDS) MUTZ-1 cells in vitro. Methods Methyl thiazolyl tetrazolium (MTT) colorimetry was used to determine the inhibitory effect of hydroxycamptothecin on the growth of human myelodysplastic syndrome MUTZ-1 cells, and the concentration inhibition curve was drawn. Flow cytometry Drug-induced apoptosis of cells; light microscopy and transmission electron microscopy of hydroxycamptothecin treatment morphological changes. Results The inhibitory effect of HCPT on MUTZ-1 cells was dose-and time-dependent. The IC50 of HCPT at different time was 91.198, 8.983, 6.204, 2.509 and 0.364, respectively. HCPT can induce MUTZ-1 cell apoptosis in a dose-dependent manner. Under light microscope, the apoptotic cells showed complete cell membrane, cell body pyknosis, nuclear pyknosis and nuclear chromatin fragmentation. Transmission electron microscopy showed apoptotic nuclear chromatin condensation, marginal set, disappearance of part of nuclear membrane, concentration of cytoplasm and density increase , The size of the irregular visible chromatin mass in the pulp. Conclusions Hydroxycamptothecin can significantly inhibit the growth of MUTZ-1 cells and induce the apoptosis of MUTZ-1 cells.
