目的 建立高效毛细管电泳法测定血浆中布比卡因的浓度。方法 取 0 .5ml血浆 ,用乙醚提取后吹干 ,用 1mmol·L-1HCl溶解 ,采用毛细管电泳法进行测定。电泳条件为 :分离用缓冲液为 2 0mmol·L-1NaH2 PO4 溶液 (pH4.8) ,实验温度 :2 5℃ ,分离电压 :30kV ,紫外检测波长 :2 0 0nm ,进样 :电动进样。结果 本法在 0 .1~ 1.0 μg·ml-1范围内呈良好的线性关系和重现性 ,方法的检测限为 0 .0 3μg·ml-1。结论 本法简便、快速、灵敏 ,为血浆中布比卡因的测定提供了新的更为理想的方法学手段
Objective To establish a high performance capillary electrophoresis method for the determination of bupivacaine in plasma. Methods: Take 0.5ml plasma, extract with ether, blow dry, dissolve with 1mmol·L-1 HCl and determine by capillary electrophoresis. The electrophoresis conditions were as follows: 20 mmol·L-1NaH2PO4 buffer (pH 4.8), temperature 25 ℃, separation voltage 30 kV, UV detection wavelength 200 nm, injection: electrokinetic injection. Results The method showed a good linearity and reproducibility in the range of 0.1-1.0 μg · ml-1. The detection limit of this method was 0. 03 μg · ml-1. Conclusion This method is simple, rapid and sensitive, and provides a new and more ideal methodological method for the determination of bupivacaine in plasma
