目的研究束缚-浸水应激(RWIS)大鼠迷走神经背核(DMV)中胆碱能神经元的活动情况。方法随机将10只雄性Wistar大鼠分为对照组和应激组,采用Fos和胆碱乙酰化酶(ChAT)免疫组织化学双标技术,统计对照组和应激组Fos、ChAT、Fos/ChAT双标阳性神经元数目。结果与对照组相比,应激组大鼠DMV大量神经元表达Fos,Fos阳性神经元主要集中于DMV尾段和吻段(P<0.01);ChAT阳性神经元主要分布于DMV中段和尾段,应激组大鼠DMV中单位面积内ChAT阳性神经元数目减少(P<0.01);Fos/ChAT双标阳性神经元的分布与Fos阳性神经元分布情况相似,RWIS组Fos/ChAT双标阳性神经元数目显著增加(P<0.01),Fos/ChAT双标阳性神经元占ChAT阳性神经元的比例在对照组和RWIS组中分别为7.17%、21.12%(P<0.01)。结论 DMV中胆碱能神经元参与RWIS调控过程。
Objective To investigate the activity of cholinergic neurons in vagal dorsal nucleus (DMV) of restraint-flooding stress (RWIS) rats. Methods Ten male Wistar rats were randomly divided into control group and stress group. Fos, ChAT and Fos / ChAT were detected by Fos and ChAT immunohistochemistry Double-labeled positive neurons. Results Compared with the control group, the expression of Fos and Fos positive neurons in the DMV neurons in stress rats were mainly concentrated in the tail and kiss segments of DMV (P <0.01). ChAT positive neurons mainly distributed in the middle and tail segments of DMV (P <0.01). The distribution of Fos / ChAT double positive neurons was similar to the distribution of Fos positive neurons in RW group. The double positive of Fos / ChAT in RWIS group (P <0.01). The ratio of Fos / ChAT double positive neurons to ChAT positive neurons in control group and RWIS group was 7.17% and 21.12% respectively (P <0.01). Conclusions Cholinergic neurons in DMV participate in the regulation of RWIS.
