目的:研究胶原纤维吸附材料对虎杖中鞣质的选择性去除作用,以及对鞣质的吸附和解吸性能。方法:以皮胶原纤维为原料,通过戊二醛交联反应制备吸附材料。将此吸附材料用于选择性去除虎杖提取物中的鞣质,并用明胶-浊度法、明胶-紫外分光法和HPLC进行分析检测。采用不同浓度的乙醇水溶液对吸附的鞣质进行静态洗脱试验,确定洗脱剂的浓度。在此基础上,在固定床中进行动态吸附和解吸试验,并采用明胶-浊度法对鞣质的去除状况进行检测。结果:所制备的胶原纤维吸附材料对鞣质具有较高的吸附选择性,可以完全除去虎杖提取物中的鞣质,而有效成分(白藜芦醇)基本不损失;该吸附材料吸附鞣质后可用30%乙醇水溶液解吸,解吸后可重复使用。结论:胶原纤维吸附材料可用于虎杖提取物中鞣质的高选择性脱除。
Objective: To study the selective removal of tannin from Polygonum cuspidatum by collagen fibers and the adsorption and desorption of tannin. Methods: Dermal fibrils were used as raw materials to prepare adsorbents by glutaraldehyde cross-linking reaction. The adsorbent material was used to selectively remove the tannin from Polygonum cuspidatum extract and analyzed by gelatin-turbidimetry, gelatin-UV spectrophotometry and HPLC. The static elution test of the tannin adsorbed by different concentrations of ethanol aqueous solution to determine the concentration of the eluent. On this basis, dynamic adsorption and desorption experiments were carried out in a fixed bed, and the removal of tannin was detected by gelatin-turbidity method. Results: The prepared collagen fiber adsorbent has high adsorption selectivity to tannin, which can completely remove the tannin in Polygonum cuspidatum extract, while the active ingredient (resveratrol) is basically not lost; the adsorbent adsorbs tannin After 30% aqueous ethanol can be desorbed, desorption can be reused. Conclusion: The collagen fiber adsorbent can be used for the high selective removal of tannin from Polygonum cuspidatum extract.
