“814”体外杀精剂以下简称“814”是从茶叶中提炼出来的外用避孕药。经药物毒性实验和其他生物学实验表明“814”较之目前国内其他的外用避孕药、具有毒性低,付作用小,杀精效果明显等优点。本文用扫描电镜和透射电镜观察经“814”作用后精子的形态学改变。材料与方法:将新鲜精液加等量的5%葡萄糖生理盐水稀释并分成四个实验组和一个对照组。四个实验组再加等量的“814”生理盐水溶液,使“814”的最终浓度分别为3%、0.3%、0.03%、0.003%。对照组以生理盐水代替之,混合后1Omin,即行标本制作。扫描电镜样品经戌二醛、锇酸固定,CO_2临界点干燥,离子溅射喷金后,行扫描电镜观察。透射电镜经双重固定后Spurr′s树脂包埋、超薄切片,行透射
“814” in vitro spermicide hereinafter referred to as “814” is extracted from tea topical contraceptives. The drug toxicity experiments and other biological experiments show that “814” compared with other domestic topical contraceptives, with low toxicity, pay a small effect, spermicidal effect and so on. In this paper, the morphological changes of spermatozoa after “814” treatment were observed by scanning electron microscopy and transmission electron microscopy. Materials and Methods: Fresh semen was diluted with 5% dextrose in saline and divided into four experimental groups and one control group. Four experimental groups plus an equal amount of “814” saline solution, so that the final concentration of “814” were 3%, 0.3%, 0.03%, 0.003%. The control group to replace the normal saline, 1Omin after mixing, that is, line specimens. Scanning electron microscopy samples were observed by scanning electron microscopy (SEM) after being oxidized by osmium tetroxide, osmium tetroxide, and dried at the critical point of CO_2. Transmission electron microscopy after double fixation Spurr’s resin embedded, ultra-thin sections, line transmission
