目的对几种常用的冷水溶性凝胶剂进行筛选及优化,得到一种适合作为快速检测纸片载体的凝胶剂混合物。方法对黄原胶、卡拉胶、瓜尔胶、聚丙烯酸钠、羟丙基甲基纤维素的凝胶性质进行筛选。选取两种适宜的凝胶进行粒径、比例的优化,确定纸片的大小,对细菌在凝胶剂载体和琼脂平板上的生长效果进行对比。结果黄原胶和聚丙烯酸钠的粒径均为0.125mm,比例为1∶5时凝胶剂具有最佳的检测效果。凝胶剂用量为0.18g,纸片的大小应为7.5cm×8cm。结论该凝胶剂混合物可以用于细菌快速检测纸片的制作。
OBJECTIVE To screen and optimize several commonly used cold water-soluble gelling agents and to obtain a gelling agent mixture suitable for rapid detection of paper supports. Methods The gel properties of xanthan gum, carrageenan, guar gum, sodium polyacrylate and hydroxypropylmethylcellulose were screened. Two suitable gels were selected for particle size and ratio optimization to determine the size of the paper and the growth of the bacteria on gel carrier and agar plates. Results The particle size of xanthan gum and sodium polyacrylate were both 0.125mm. The gel was the best one when the ratio was 1: 5. Gel amount of 0.18g, the size of the paper should be 7.5cm × 8cm. Conclusion The gel mixture can be used for bacterial rapid detection of paper production.
