目的研究维吾尔族宫颈炎、宫颈上皮内瘤变和宫颈癌组织中极性蛋白Par3与上皮型钙黏蛋白(Ecadherin)、β-连环蛋白(β-catenin)、神经性钙黏蛋白(N-cadherin)、转录因子Smad1、波形蛋白(Vimentin)的表达及其临床病理学意义。方法收集维吾尔族妇女宫颈鳞癌(cervical squamous cell carcinoma,CSCC)、宫颈上皮内瘤变(cervical intraepithelial neoplasia,CIN)患者和正常宫颈石蜡包埋组织标本189例,采用免疫组织化学的方法分别检测Par3与E-cadherin、N-cadherin、β-catenin、Smad1及vimentin的表达情况。结果 Par3、E-cadherin、N-cadherin、β-catenin、Smad1及vimentin在正常宫颈组织、CIN和CSCC组织中呈不同程度阳性表达。正常宫颈组织中Par3、E-cadherin、N-cadherin、β-catenin、Smad1及vimentin阳性表达率分别为85.4%、53.7%、19.5%、53.7%、22.0%、26.8%。在CIN组织中阳性表达率分别为50.0%、31.3%、34.3%、9.3%、56.3%、43.8%。在宫颈癌中阳性表达率分别为17.2%、6.8%、77.6%、8.6%、76.7%、87.1%。随着宫颈病变的加重,Par3、E-cadherin、β-catenin的表达显著下调(P<0.05)。N-cadherin、Smad1及vimentin表达显著上调(P<0.05)。Par3及E-cadherin、N-cadherin、β-catenin、Smad1蛋白的表达均与宫颈癌FIGO分期、分化程度及淋巴结转移有关(P<0.05)。而Vimentin的表达则仅与浸润性宫颈癌组织分级、分化程度及淋巴结转移有关(P<0.05)。结论随着宫颈病变的进展,宫颈癌组织中Par3蛋白低表达可能参与了宫颈上皮间充质转变的过程。
Objective To investigate the relationship between Par3 and Ecadherin, β-catenin and N-cadherin in Uygur cervicitis, cervical intraepithelial neoplasia and cervical cancer. ), The expression of Smad1, vimentin and their clinicopathological significance. Methods A total of 189 Uygur women with cervical squamous cell carcinoma (CSCC), cervical intraepithelial neoplasia (CIN) and normal cervical paraffin-embedded tissue samples were collected from 189 patients. Immunohistochemistry was used to detect Par3 And E-cadherin, N-cadherin, β-catenin, Smad1 and vimentin expression. Results The positive expression of Par3, E-cadherin, N-cadherin, β-catenin, Smad1 and vimentin in normal cervical tissue, CIN and CSCC tissues were positive. The positive expression rates of Par3, E-cadherin, N-cadherin, β-catenin, Smad1 and vimentin in normal cervical tissue were 85.4%, 53.7%, 19.5%, 53.7%, 22.0% and 26.8% respectively. The positive rates in CIN tissues were 50.0%, 31.3%, 34.3%, 9.3%, 56.3% and 43.8%, respectively. The positive rates in cervical cancer were 17.2%, 6.8%, 77.6%, 8.6%, 76.7%, 87.1% respectively. With the increase of cervical lesions, the expressions of Par3, E-cadherin and β-catenin were significantly down-regulated (P <0.05). The expressions of N-cadherin, Smad1 and vimentin were significantly up-regulated (P <0.05). The expression of Par3, E-cadherin, N-cadherin, β-catenin and Smad1 in cervical cancer correlated with FIGO stage, differentiation and lymph node metastasis (P <0.05). However, the expression of Vimentin was only associated with the grade, differentiation and lymph node metastasis of invasive cervical cancer (P <0.05). Conclusion With the progress of cervical lesions, the low expression of Par3 protein in cervical cancer may be involved in the process of cervical epithelial mesenchymal transition.
