目的研究麦门冬汤合苇茎汤(简称金方/JIN)对Lewis肺癌小鼠移植瘤的抑制作用并初步探讨药物作用机制。方法皮下注射LLC(Lewis lung carcinoma,Lewis肺癌细胞)于C57/BL6小鼠右腋下,观察小鼠生存期、抑瘤率、胸腺指数和肿瘤组织EGFR/Raf/MEK的表达。结果金方可以显著延长小鼠的生存时间,与对照组相比有显著性差异(P<0.01),金方、DDP组对荷瘤小鼠的抑瘤率分别为35%、89%,金方组胸腺指数明显升高,DDP组胸腺指数明显下降,金方、DDP组的EGFR表达均下降(P>0.05,P<0.01),而各组Raf和MEK表达无显著性差异。结论金方可以显著延长荷瘤小鼠的生存期,有一定的抑制肿瘤增长和增强免疫能力的作用,其机制可能与其下调肿瘤组织中EGFR的表达进而影响其下游信号通路的传导相关。
Objective To study the inhibitory effect of Mai Men Dong Decoction (JIN) on transplanted Lewis lung carcinoma in mice and to explore the mechanism of the drug action. Methods LLC (Lewis lung carcinoma, Lewis lung cancer cells) were injected subcutaneously into the right armpit of C57 / BL6 mice to observe the survival, tumor inhibition rate, thymus index and the expression of EGFR / Raf / MEK in mice. Results Jinfang can significantly prolong the survival time of mice, compared with the control group (P <0.01), Jin Fang, DDP group tumor-bearing mice tumor-inhibitory rates were 35%, 89% The index of thymus in group A and group DDP was significantly lower than that in group DDP (P <0.05, P <0.01). The expression of Raf and MEK in group DDP had no significant difference. Conclusion Jinfang can significantly prolong the survival of tumor-bearing mice, and have a certain inhibitory effect on tumor growth and immune enhancement. The mechanism may be related to its down-regulation of EGFR expression and its influence on the downstream signaling pathway.
