目的和方法:本研究应用MIT比色法分析人源性单克隆抗体(HMcAb)AT4对于人卵巢癌细胞ao10/17的体外细胞毒。结果:HMcAbAT4能激活人和家兔血清补体介导的ao10/17细胞的细胞溶解,介导人效应细胞对于靶肿瘤细胞的抗体依赖细胞毒,增强人LAK细胞对于ao10/17细胞的杀伤作用。结论:HMcAbAT4在体外可介导对于人卵巢癌细胞的抗体依赖效应细胞的细胞毒和补体依赖细胞溶解,因而可能成为卵巢癌免疫治疗研究特别有希望的试剂。
PURPOSE AND METHODS: In this study, the cytotoxicity of human monoclonal antibody (HMcAb) AT4 on human ovarian cancer cell line ao10 / 17 was analyzed by MIT colorimetric assay. Results: HMcAbAT4 could activate human luteal and rabbit serum-mediated cell lysis of ao10 / 17 cells and mediate antibody-dependent cytotoxicity of human effector cells against target tumor cells and enhance the killing effect of human LAK cells on ao10 / 17 cells. CONCLUSION: HMcAbAT4 can mediate the cytotoxicity and complement-dependent cell lysis of antibody-dependent effector cells in human ovarian cancer cells in vitro and thus may be a particularly promising reagent for immunotherapy of ovarian cancer.
