麻疯树油质蛋白家族基因的鉴定与进化分析

油质蛋白(OLE)是位于油体表面的一类膜结合蛋白,这类蛋白在油体形成过程中起重要作用。利用生物信息学手段在麻疯树基因组中共鉴定出6个JcOLE基因,并详细剖析这些基因的序列与进化特征。系统进化分析显示:JcOLE基因被分为5个类群,并在进化树末端发现了2对同源基因。序列分析表明:6个OLE基因具有3种基因结构类型,同时6个OLE蛋白具有3种跨膜结构与保守基序组成模式,共享了同一个典型OLE功能结构域。选择压力检测显示,JcOLE5和JcOLE6同源基因对受到负选择作用,但JcOLE1和JcOLE2同源基因对受到正选择作用,发生了适应性进化。EST数目与来源分析显示:除JcOLE4外,麻疯树OLE基因在胚中表达;JcOLE4在花中表达,而JcOLE3和JcOLE4在种子中表达。这些研究有助于进一步解析麻疯树OLE基因的生物学功能。 Oleosin (OLE) is a type of membrane-bound protein located on the surface of oil bodies. These proteins play an important role in the formation of oil bodies. Six JcOLE genes were identified in Jatropha curcas genome using bioinformatics methods, and the sequence and evolution characteristics of these genes were analyzed in detail. Phylogenetic analysis showed that JcOLE genes were divided into five groups and two pairs of homologous genes were found at the end of the phylogenetic tree. Sequence analysis showed that the six OLE genes have three kinds of gene structure types, and six OLE proteins have three transmembrane structure and conservative motif composition pattern, sharing the same typical OLE functional domain. Selective pressure detection showed that the JcOLE5 and JcOLE6 homologues were negatively selected, but the JcOLE1 and JcOLE2 homologues were positively selected and adaptively evolved. EST number and source analysis showed that, except JcOLE4, Jatropha curcas OLE gene was expressed in the embryo; JcOLE4 was expressed in the flower, while JcOLE3 and JcOLE4 were expressed in the seed. These studies will help further elucidate the biological function of Jatropha curcas OLE gene.
