目的:探讨苯丙胺对额叶皮质部位AKT信号通路上游信号分子的影响。方法:选取健康雄性成年SD大鼠50只,分为空白组、生理盐水组和苯丙胺(AMPH)组。用旷场(open field)系统测试大鼠的行为学变化,用Western blot分析AKT信号通路上游信号分子的变化,用免疫组织化学观察相应分子阳性细胞的变化。结果:注射苯丙胺后大鼠活动度增加,且出现刻板行为。AMPH组大鼠额叶皮质pm TOR含量比空白组及生理盐水组减少,且pm TOR阳性细胞的数量也相应减少,而p PP2A和PHLPP没有显著性变化。结论:苯丙胺抑制AKT信号通路可能与减少pm TOR的表达密切相关。
Objective: To investigate the effect of amphetamine on the signal molecules upstream of AKT signaling pathway in frontal cortex. Methods: Fifty adult male Sprague-Dawley rats were randomly divided into blank group, normal saline group and amphetamine group (AMPH). The behavior of the rats was tested by open field system. The changes of signal molecules in the upstream of AKT signaling pathway were analyzed by Western blot. The changes of the corresponding positive cells were observed by immunohistochemistry. Results: After injection of amphetamine, the activity of rats increased and their stereotyped behaviors appeared. The pm TOR content in frontal cortex of rats in AMPH group decreased compared with that of blank group and normal saline group, and the number of pm TOR positive cells decreased accordingly, while there was no significant change in p PP2A and PHLPP. Conclusion: Amphetamine inhibit AKT signaling pathway may be closely related to the reduction of pm TOR expression.
