EB病毒潜伏膜蛋白(LMP)具有致癌活性,构建含金属硫蛋白基因-1(MT-1)调控区和LMP基因的pBR322-MT-LMP质粒,用显微注射法将MT-LMP转基因接种于小鼠受精卵内制备转基因小鼠的动物模型。结果:a.注射后卵的存活率和仔鼠出生率分别为73%和13%;b.转基因鼠尾部组织DNA分析,PCR法证明二只小鼠有LMP基因扩增带,Southern杂交亦显示PCR阳性鼠有特异的LMP杂交信号,这说明LMP基因已整合于鼠基因组,整合率为8%。结果说明,构建携带LMP基因的转基因动物模型已成功。
EB virus latent membrane protein (LMP) has carcinogenic activity, and pBR322-MT-LMP plasmid containing the metallothionein gene-1 (MT-1) regulatory region and LMP gene was constructed, and the MT-LMP transgene was inoculated by microinjection. An animal model of transgenic mice was prepared in mouse fertilized eggs. Result: a. After the injection, the survival rate of the eggs and the birth rate of the offspring were 73% and 13%, respectively; b. DNA analysis of the tail of transgenic mice showed that the two mice had LMP gene amplification bands. Southern hybridization also showed that the PCR positive mice had specific LMP hybridization signals, indicating that the LMP gene has been integrated in the mouse genome, and the integration rate was 8%. . The results show that the construction of a transgenic animal model carrying the LMP gene has been successful.
