目的研究甲状腺癌组织中核因子κB(NF-κB)和信号转导子与转录激活子3(STAT3)的表达情况。方法选取69例甲状腺癌和58例正常甲状腺组织标本,荧光定量PCR检测NF-κB以及STAT3 mRNA水平,ELISA检测其组织匀浆中NF-κB和STAT3蛋白含量、免疫组织化学检测NF-κB和STAT3定位。结果与正常甲状腺组织相比,甲状腺癌组织中NF-κB和STAT3的mRNA和蛋白表达水平显著升高,免疫组织化学结果显示,甲状腺癌组织中STAT3阳性率(89.3%)显著高于正常甲状腺组织(10.2%)。结论 STAT3介导的NF-κB信号通路在甲状腺癌患者的组织活性增强。
Objective To investigate the expression of nuclear factor-κB (NF-κB) and signal transducer and activator of transcription 3 (STAT3) in thyroid cancer. Methods Sixty-nine thyroid carcinomas and 58 normal thyroid tissue samples were collected. The levels of NF-κB and STAT3 mRNA were detected by real-time quantitative PCR. The levels of NF-κB and STAT3 were detected by ELISA. The expressions of NF-κB and STAT3 were detected by immunohistochemistry Positioning. Results Compared with normal thyroid tissue, the mRNA and protein expressions of NF-κB and STAT3 were significantly increased in thyroid cancer tissues. Immunohistochemistry showed that the positive rate of STAT3 (89.3%) in thyroid cancer tissues was significantly higher than that in normal thyroid tissues (10.2%). Conclusion STAT3-mediated NF-κB signaling pathway in thyroid cancer patients with increased tissue activity.
