目的:研究含金属硫蛋白(MT)蛋奶粉对小鼠电离辐射损伤的保护作用。方法:小鼠受2.5Gy(12.5mGy·min-1)X射线照射后,灌胃给予含MT蛋奶粉,连续14d后检测外周血白细胞数、淋巴细胞增殖率、股骨骨髓细胞DNA含量、胸腺细胞周期进程以及肝脏和肾脏丙二醛(MDA)含量和抗氧化酶(SOD、CAT和GSHPx)活性。结果:受照射小鼠给予含MT蛋奶粉后,尤其是含中高剂量MT蛋奶粉组,白细胞数、淋巴细胞增殖率和股骨骨髓细胞DNA含量明显高于辐射对照组(P<0.01);含MT蛋奶粉加照射组胸腺细胞G0/G1期细胞数明显低于辐射对照组(P<0.01),S期和G2/M期细胞数明显高于辐射对照组(P<0.01或P<0.05);照射组小鼠肝脏和肾脏MDA含量较正常对照组显著增加(P<0.01或P<0.05),抗氧化酶SOD、CAT和GSHPx活性则较正常对照组显著降低(P<0.01或P<0.05),而含MT蛋奶粉加照射组与辐射对照组比较,肝脏和肾脏MDA水平明显降低(P<0.01),抗氧化酶SOD、CAT和GSHPx活性显著增高(P<0.01或P<0.05)。结论:含MT蛋奶粉可明显改善X射线照射对小鼠的损伤效应,具有保护作用。
Objective: To study the protective effect of metallothionein (MT) milk powder on ionizing radiation injury in mice. Methods: Mice were treated with 2.5Gy (12.5mGy · min-1) X-ray and intramuscularly given milk powder containing MT. The number of peripheral blood leukocytes, lymphocyte proliferation rate, DNA content of femur bone marrow cells, Cycle progression and liver and kidney malondialdehyde (MDA) content and antioxidant enzymes (SOD, CAT and GSHPx) activity. Results: The number of leucocyte, lymphocyte proliferation and the content of DNA in femur marrow cells were significantly higher than those in irradiated control group (P <0.01) after irradiated mice were given MT milk powder, The number of G0 / G1 phase cells in irradiated group and irradiated group was significantly lower than that in irradiated control group (P <0.01). The number of cells in S phase and G2 / M phase was significantly higher than that in irradiated control group (P <0.01 or P <0.05). Compared with normal control group, the content of MDA in liver and kidney of irradiation group was significantly increased (P <0.01 or P <0.05), and the activities of SOD, CAT and GSHPx were significantly decreased (P <0.01 or P <0.05) (P <0.01), and the activities of SOD, CAT and GSHPx were significantly increased (P <0.01 or P <0.05) compared with the radiation control group. Conclusion: Milk powder containing MT can obviously improve the damage effect of X-ray irradiation on mice and has protective effect.
