目的:研究丁咯地尔和α-硫辛酸(ALA)对糖尿病(DM)大鼠肾功能的保护作用。方法:雄性SD大鼠用链脲佐菌素(60 mg/kg)腹腔注射诱导DM。大鼠随机分为健康组(NC)、DM组(DMC)、DM+ALA组(ALA组)、DM+丁咯地尔组(丁咯地尔组)。ALA组和丁咯地尔组每天给予ALA 100 mg/kg灌胃和丁咯地尔(0.1 g/kg)腹腔注射,NC组和DMC组大鼠给予等量生理盐水每日灌胃。干预4周后,测观察治疗后大鼠血糖、尿素氮、血肌苷,内生肌苷清除率、24小时尿蛋白排泄率,比色法检测血清抗氧化酶和丙二醛(MDA)含量。结果:①丁咯地尔和α-硫辛酸降低糖尿病大鼠的尿素氮、血肌苷、24h尿白蛋白排泄率、丙二醛,增加超氧化物歧化酶②两种药物联合用药具有协同作用。结论:丁咯地尔和ALA可能通过抑制机体氧化应激水平,减低早期DM大鼠肾脏损害。
Objective: To study the protective effects of buflomedil and α-lipoic acid (ALA) on renal function in diabetic rats. Methods: Male Sprague-Dawley rats were induced with streptozotocin (60 mg / kg) intraperitoneally to induce DM. Rats were randomly divided into healthy group (NC), DM group (DMC), DM + ALA group (ALA group) and DM + buflomedil group (buflomedil group). The ALA group and the buflomedil group were intraperitoneally injected with 100 mg / kg ALA and buprenorphine (0.1 g / kg). The rats in NC group and DMC group were given intragastric administration of normal saline daily. After intervention for 4 weeks, the blood glucose, urea nitrogen, creatinine, endogenous creatinine clearance rate and 24-hour urinary protein excretion rate were measured and the levels of serum antioxidant enzymes and malondialdehyde (MDA) were measured by colorimetry. . Results: (1) Buflomedil and α-lipoic acid can reduce the excretion of urinary nitrogen, serum creatinine and urinary albumin excretion rate, and the synergistic effect of the two drugs in combination with malondialdehyde and superoxide dismutase . CONCLUSION: Buflomedil and ALA may reduce renal oxidative damage in early DM rats by inhibiting oxidative stress.
