目的探讨Wnt5a基因及蛋白在糖尿病大鼠肾组织中的表达及意义。方法链脲佐菌素(streptozotocin,STZ)诱导大鼠糖尿病模型,分为2、4、8、12、16、24周组,每一时间组均设正常对照,测定各组血糖、24 h尿蛋白、血肌酐水平。免疫组化方法检测Wnt5a及FN蛋白表达,蛋白免疫印迹(western blotting)检测Wnt5a蛋白表达,逆转录-聚合酶链反应检测Wnt5a及FN基因表达。结果与正常对照组比较,糖尿病组Wnt5a蛋白表达分别增多了65.5%、72.2%、60%、68.8%、68.8%及63.0%(P<0.01);糖尿病各组Wnt5a和FN mRNA的表达分别增多了50.9%、50%、35.7%、50.9%、55%、54.2%和35.5%、64.6%、34.1%、61.8%、66.7%、64.6%(P<0.01);Wnt5a与FN mRNA表达水平呈正相关(r=0.714,P<0.01)。结论 Wnt5a蛋白和基因在糖尿病大鼠肾组织表达增多,Wnt5a可能参与了糖尿病的发生发展过程。
Objective To investigate the expression and significance of Wnt5a gene and protein in the kidney of diabetic rats. Methods Diabetic rats were induced by streptozotocin (STZ) and divided into groups of 2, 4, 8, 12, 16, and 24 weeks. Each group was given normal control. Blood glucose, Protein, serum creatinine levels. The expression of Wnt5a and FN was detected by immunohistochemistry, the expression of Wnt5a was detected by western blotting, and the expression of Wnt5a and FN was detected by reverse transcription polymerase chain reaction (RT-PCR). Results Compared with the normal control group, the expression of Wnt5a in diabetic group increased by 65.5%, 72.2%, 60%, 68.8%, 68.8% and 63.0%, respectively (P <0.01) 50.9%, 50%, 35.7%, 50.9%, 55%, 54.2% and 35.5%, 64.6%, 34.1%, 61.8%, 66.7% and 64.6% respectively (P <0.01) .Wnt5a and FN mRNA expression levels were positively correlated r = 0.714, P <0.01). Conclusion Wnt5a protein and gene expression increased in the kidney of diabetic rats, Wnt5a may be involved in the development of diabetes.
