目的分析和预测猪带绦虫腺苷酸激酶(ADK)基因及其编码的蛋白结构和特性。方法利用各种在线生物信息网站及其它分析软件包,分析从猪带绦虫成虫cDNA质粒文库中识别的腺苷酸激酶基因的结构和编码区序列,分析、预测编码的蛋白质的理化特性、抗原表位、翻译后的修饰、功能域、亚细胞定位、拓扑结构、二级结构、三维空间构象等。结果该基因全长864 bp,编码区为110~758,编码215个氨基酸,为全长基因。理论分子量、等电点分别为23771.4 Da和6.86;没有跨膜区;三维结构建模图显示其线性表位的部位。结论应用生物信息方法从猪带绦虫成虫cDNA文库中筛选出了腺苷酸激酶基因的cDNA全长序列并预测得到其结构与功能方面的信息。
Objective To analyze and predict the adenovirus kinase (ADK) gene of Taenia solium and its encoded protein structure and characteristics. Methods Various online bioinformatics websites and other analysis software packages were used to analyze the structure and coding region of adenylate kinase gene identified from the cDNA plasmid library of adult Taenia solium. The physical and chemical properties of the encoded protein were predicted and analyzed. Position, post-translational modification, functional domain, subcellular localization, topology, secondary structure, three-dimensional conformation and so on. Results The full-length gene was 864 bp in length and coding region was 110-758, encoding 215 amino acids. The theoretical molecular weight, isoelectric point were 23771.4 Da and 6.86 respectively; there was no transmembrane region; and the three-dimensional structural modeling showed the site of its linear epitope. Conclusion The full-length cDNA sequence of adenylate kinase gene was screened from cDNA library of adult Taenia solium using bioinformatics method and its structure and function were predicted.
