【目的】研究枳实白芍药对配伍高效液相色谱(HPLC)图谱中化学成分变化及与药效的相关性。【方法】采用冰醋酸灌肠与束缚夹尾相结合方法复制腹泻型肠易激综合征(D-IBS)大鼠模型,以小肠推进率、24 h粪点数和腹部回缩反射评分作为药效指标,并设计枳实白芍药对不同配伍组(质量比为1∶1、2∶1、1∶2),进行HPLC测定,将HPLC图谱中11个色谱峰与药效指标进行灰色关联度相关性分析。【结果】依据灰色关联度大小,确定各成分对小肠推进率贡献大小顺序:8>11>9>10>2>7>1>5>3;对24 h粪点数贡献大小顺序:8>9>5>11>7>10>1>2>3;对大鼠腹部回缩反射评分贡献大小:10>5>11>8>9>7>1>2>3。其中9号峰为柚皮苷、10号峰为橙皮苷、11号峰为新橙皮苷。【结论】枳实白芍药对可改善D-IBS大鼠胃肠道情况,与其所含的多种化学成分相关。
【OBJECTIVE】 To study the relationship between the changes of chemical composition and the pharmacodynamics in compatibility HPLC chromatogram of Poncirus trifoliate white peony. 【Method】 The diarrhea-predominant irritable bowel syndrome (D-IBS) rat model was duplicated by the combination of glacial acetic acid enema and restraint pinch tail. The small intestine propulsion rate, 24 h fecal point and abdominal retraction reflex score were taken as the pharmacodynamic parameters , And the different fractions of Citrus aurantium peony (compatibility ratio 1: 1, 2: 1, 1: 2) were designed and measured by HPLC. The 11 chromatographic peaks in HPLC were correlated with the pharmacodynamics indexes analysis. 【Results】 According to the degree of gray relational degree, the order of contribution of each component to small intestine propulsion rate was determined: 8> 11> 9> 10> 2> 7> 1> 5> 3; > 5> 11> 7> 10> 1> 2> 3; The contribution to the rat reflex reflex score: 10> 5> 11> 8> 9> 7> 1> 2> 3. Among them, Peak 9 was naringin, Peak 10 was hesperidin and Peak 11 was neohesperidin. 【Conclusion】 Citrus aurantium Paeoniae Alba is able to improve gastrointestinal conditions in D-IBS rats, which is related to the various chemical components it contains.
