目的探讨氧化型低密度脂蛋白(ox-LDL)及荷叶生物碱对THP-1源性巨噬细胞中血凝素样氧化型低密度脂蛋白受体1(LOX-1)表达的影响。方法①用不同浓度(0、25、50、100mg/L)ox-LDL刺激人单核细胞系THP-1源性巨噬细胞;②预先用不同浓度(0、25、50、100mg/L)荷叶生物碱处理,再予50mg/Lox-LDL培养;荧光实时定量PCR和Westernblot检测各组细胞LOX-1mRNA和蛋白的表达。结果 ox-LDL各剂量组皆可上调LOX-1mRNA及蛋白水平,在0~50mg/L剂量范围内呈明显的剂量-效应关系(P<0.01)。荷叶生物碱处理组细胞LOX-1mRNA及蛋白水平呈浓度依赖性降低(P<0.01)。结论 ox-LDL可上调THP-1源性巨噬细胞LOX-1的表达;荷叶生物碱可抑制ox-LDL诱导的THP-1源性巨噬细胞LOX-1的表达。
Objective To investigate the effect of oxidized low density lipoprotein (ox-LDL) and lotus leaf alkaloids on the expression of LOX-1 in THP-1-derived macrophages. Methods ① THP-1-derived macrophages were stimulated with different concentrations of ox-LDL (0, 25, 50 and 100 mg / L) Lotus leaf alkaloids, and then 50mg / Lox-LDL culture; real-time quantitative PCR and Western blot detection of LOX-1 mRNA and protein expression in each group. Results Ox-LDL could up-regulate the mRNA and protein levels of LOX-1 in all dose groups. The dose-effect relationship was significant at the dose range of 0 ~ 50 mg / L (P <0.01). LOX-1 mRNA and protein levels in lotus leaf alkaloids decreased in a concentration-dependent manner (P <0.01). Conclusion Ox-LDL can up-regulate the expression of LOX-1 in THP-1-derived macrophages. Lotus leaf alkaloids can inhibit the expression of LOX-1 in THP-1-derived macrophages induced by ox-LDL.
