内生真菌已经成为发现结构新颖、活性显著的次级代谢产物的重要来源,红树林内生真菌的代谢产物中发现了许多结构多样的化合物,引起了人们的广泛重视。采用硅胶柱色谱、Sephadex LH-20凝胶柱色谱、以及制备薄层色谱等方法从南海红树林植物桐花树内生杂色拟盘多毛孢真菌大米发酵产物中的乙酸乙酯提取物中得到4个代谢产物,其中1个新氯代二苯甲酮衍生物pestalachloride E(2),2个已知的二苯甲酮衍生物pestalachlorides C(1)和B(3),1个已知的蒽醌化合物,大黄素甲醚physcion(4)。新化合物通过一维和二维核磁共振谱鉴定,并通过高分辨电喷雾质谱确认。细胞毒实验结果表明化合物1显示良好的抗肿瘤活性,IC50在2.8~82.4μmol/L区间。化合物3显示具有抗炎活性,对环氧合酶-2的抑制活性为IC50 2.4μmol/L。
Endophytic fungi have become important sources for discovering novel and highly active secondary metabolites. Many structurally diverse compounds have been found in the metabolites of mangrove endophytic fungi, causing widespread concern. Silica gel column chromatography, Sephadex LH-20 gel column chromatography, and preparative thin layer chromatography were used to obtain the ethyl acetate extract from the rice fermentation product of the endophytic A. polypora in the South China Sea mangrove plant Four metabolites, one of the new chlorobenzophenone derivatives pestalachloride E (2), two known benzophenone derivatives pestalachlorides C (1) and B (3), one known Anthraquinone compounds, physcion of physcion (4). The new compounds were identified by one-dimensional and two-dimensional nuclear magnetic resonance spectroscopy and confirmed by high-resolution electrospray mass spectrometry. The result of cytotoxicity showed that compound 1 showed good anti-tumor activity with IC50 of 2.8 ~ 82.4μmol / L. Compound 3 showed anti-inflammatory activity with an inhibitory activity against cyclooxygenase-2 of IC50 2.4 [mu] mol / L.
