目的建立薄层色谱法对茯苓山药片中非法添加化学药品格列本脲进行快速筛查。方法以格列本脲对照品作对照,以三氯甲烷-甲醇(1:1)为溶剂进行提取,硅胶G薄层板上点样,以三氯甲烷-甲醇-甲酸(15:10:3)为展开剂展开,紫外灯光(365 nm)下检视荧光斑点。结果采用薄层色谱法能够高效快速地排查茯苓山药片中是否非法添加化学药品格列本脲,在与格列本脲对照品色谱相应的位置上,未显相同颜色的荧光斑点为阴性(样品不含格列本脲),显示相同颜色的荧光斑点为阳性(样品含格列本脲)。结论所建立的方法操作简便,且稳定性和重现性好,结果比较准确,适用于对茯苓山药片中的格列本脲的快速筛查。
OBJECTIVE To establish a thin-layer chromatography for rapid screening of glibenclamide illegally added in the Poria yam tablets. Methods Glibenclamide reference substance was compared with chloroform - methanol (1: 1) as the solvent, and then spotted on silica gel G plate with chloroform - methanol - formic acid (15: 10: 3 ) For the developer to start, under UV light (365 nm) view fluorescent spots. Results Thin-layer chromatography (TLC) was used to detect whether glibenclamide was illegally added into the Tuckahoe tablets. The spots of the same color were not found to be negative at the corresponding position of glyburide reference substance (sample Free from glibenclamide) showing a positive fluorescent spot of the same color (sample containing glyburide). Conclusion The established method is simple and easy to operate with good stability and reproducibility and accurate results. It is suitable for the rapid screening of glibenclamide in Poria cocos tablets.
