目的:探讨基质金属蛋白酶2(MMP2)及组织金属蛋白酶抑制剂2(TIMP2)与大肠癌的关系。方法:采用RTPCR方法对12例结肠癌、20例直肠癌(其中4例为复发癌)组织及相应正常组织中MMP2和TIMP2mRNA表达水平进行测定。结果:27/32的肿瘤组织和18/32的正常组织中有MMP2mRNA表达,TIMP2mRNA的表达比例分别为29/32和25/32。MMP2在大肠癌组织中的表达水平与患者年龄呈负相关。11例正常组织MMP2mRNA水平高于肿瘤组织水平的病例中,结肠癌的比例明显高于直肠癌(18/32,16/32,P<0.05)。结论:大肠癌不同年龄所表现的不同生物学行为可能与MMP2在肿瘤组织中的表达水平有关。结肠癌与直肠癌具有不同的生物学行为
Objective: To investigate the relationship between matrix metalloproteinase 2 (MMP 2) and tissue inhibitor of metalloproteinase 2 (TIMP 2) and colorectal cancer. Methods: The expression levels of MMP-2 and TIMP-2 mRNA in 12 cases of colon cancer, 20 cases of rectal cancer (including 4 cases of recurrence) and corresponding normal tissues were determined by RT-PCR. RESULTS: MMP-2 mRNA was expressed in 27/32 tumor tissues and 18/32 normal tissues, and the expression ratio of TIMP-2 mRNA was 29/32 and 25/32, respectively. The expression level of MMP 2 in colorectal cancer was negatively correlated with the age of the patients. In 11 cases of normal tissue MMP 2 mRNA levels higher than the level of tumor tissue, the proportion of colon cancer was significantly higher than rectal cancer (18/32, 16/32, P <0.05). Conclusion: The different biological behaviors of colorectal cancer at different ages may be related to the expression level of MMP 2 in tumor tissues.
