近年来扫描电镜的细胞表面分子标记术有较大进展,使用的标记物已达十几种,这些标记物具有易识别、稳定性好等特点。使用的配体则有植物凝集素、抗体和药物三类。通常标记物与配体的连接采用两种方法,即采用交联剂的共价键法及非共价键吸附法。标记物-配体复合物可采用层析、离心等方法分离纯化,然后通过各种方法与细胞表面的特异分子结合,从而在扫描电镜下观察、分析这些表面分子的数量和分布。本文对该技术所涉及的实验方法及注意事项作了较详尽的讨论。
In recent years, scanning electron microscopy of cell surface molecular markers have made great progress, the use of markers has reached a dozen, these markers have easy identification, good stability and so on. The ligands used are lectins, antibodies and drugs three categories. Usually labels and ligands using two methods, namely, the use of cross-linking agent covalent and non-covalent adsorption method. The label-ligand complex can be isolated and purified by chromatography, centrifugation and the like, and then bound to specific molecules on the cell surface through various methods to observe and analyze the number and distribution of these surface molecules under a scanning electron microscope. This article discusses the experimental methods and precautions involved in this technology in more detail.
