目的研究枸杞多糖(LBP)对THP-1单核细胞源性巨噬细胞内内质网应激反应蛋白及氧化低密度脂蛋白(ox-LDL)含量变化的影响。方法培养THP-1单核细胞,加入佛波酯(phorbol 12-myristate 13-acetat,PMA)诱导分化成THP-1巨噬细胞,随后,加入不同浓度的LBP(20?g/ml、50?g/ml、100?g/ml、200?g/ml)孵育24h后收集细胞。用酶联免疫吸附法(E LISA)检测细胞上清液中ox-LDL及内质网应激反应蛋白(Grp7 8,CHOP,Xbp-1)的含量变化。结果不同浓度的LBP孵育THP-1巨噬细胞,细胞中的ox-LDL与内质网应激反应蛋白的含量均降低。结论 LBP可以降低ox-LDL及内质网应激反应蛋白的含量,对预防动脉粥样硬化的发生发展起到一定的作用。
Objective To investigate the effects of LBP on the changes of endoplasmic reticulum stress protein and oxidized low density lipoprotein (ox-LDL) in THP-1 monocyte-derived macrophages. Methods THP-1 monocytes were cultured and induced to differentiate into THP-1 macrophages by addition of phorbol 12-myristate 13-acetat (PMA). Subsequently, different concentrations of LBP (20 μg / ml, g / ml, 100? g / ml, 200? g / ml). The changes of ox-LDL and ER stress protein (Grp7 8, CHOP, Xbp-1) in the cell supernatant were detected by enzyme-linked immunosorbent assay (ELISA). Results THP-1 macrophages were incubated with different concentrations of LBP. The content of ox-LDL and ER in the cells decreased. Conclusion LBP can reduce the content of ox-LDL and endoplasmic reticulum stress protein, which play a role in preventing the occurrence and development of atherosclerosis.
