目的 探讨金黄色葡萄球菌肠毒素B(SEB)对小鼠树突状细胞 (DC)表面I-Eκ 分子表达的影响。方法 密度梯度离心获得小鼠脾单个核细胞 ,Panning法纯化非贴壁细胞得到DC ;免疫组化SABC法检测DC纯度 ;流式细胞术检测SEB刺激后DC表面I -Eκ 分子的表达。结果 DC纯度为 70 %~ 80 %。I -Eκ 的表达在 10 0ng/mlSEB刺激 8h后达到高峰。结论 SEB可显著提高DC表面I -Eκ 分子的表达 ,该刺激作用有一最佳量效和时效关系。
Objective To investigate the effect of Staphylococcus aureus enterotoxin B (SEB) on the expression of I-Eκ on the surface of mouse dendritic cells. Methods The spleen mononuclear cells were obtained by density gradient centrifugation. DCs were purified by non-adherent cells by Panning method. The purity of DCs was detected by immunohistochemical SABC method. The expression of I-E molecules on DCs was detected by flow cytometry. Results DC purity of 70% to 80%. The expression of I-Eκ peaked at 10 ng / ml SEB for 8 h. Conclusion SEB can significantly increase the expression of I-Eκ on the surface of DC, and this stimulation has an optimal dose-and-time relationship.
