目的:探讨不同剂量浓缩牛肉汤对局灶性脑缺血大鼠脾脏IL-4和TNF-α表达的影响。方法:将60只雄性SD大鼠随机分入组I(假手术组I)、组II(假手术组II)、组III(模型组)、组IV[低剂量浓缩牛肉汤组200 mg/(kg·d)]、组V[中剂量浓缩牛肉汤组400 mg/(kg·d)]和组VI[高剂量浓缩牛肉汤组600 mg/(kg·d)],每组各10只。组IV~组VI大鼠在造模后分别以200、400、600 mg/(kg·d)浓缩牛肉汤灌胃,组II予600mg/(kg·d)浓缩牛肉汤灌胃,组I和组III以等量生理盐水灌胃。于术后2 h和7 d分别进行大鼠神经功能缺损评分,于术后7 d测量脾质量指数和脑梗死体积,并用酶联免疫吸附测定(ELISA)法检测脾脏TNF-α和IL-4水平。结果:术后2 h和7 d,与组III相比,组V和组VI大鼠神经功能缺损评分下降有统计学意义(P<0.05);术后7 d,组I和组II大鼠脾指数、脾脏IL-4和TNF-α表达无差异;与组III相比,组V和组VI大鼠脾指数上升,脑梗死体积减少,TNF-α表达水平降低,组VI大鼠脾脏IL-4表达显著上升(P<0.05)。结论:浓缩牛肉汤可改善大鼠神经功能缺损,减少脑梗死体积,升高脑缺血大鼠脾指数,降低脾脏TNF-α水平,升高脾脏IL-4表达,且呈剂量依赖性。
Objective: To investigate the effect of different doses of concentrated beef broth on the expression of IL-4 and TNF-α in spleen of focal cerebral ischemia rats. Methods: Sixty male Sprague-Dawley rats were randomly divided into three groups: sham operation group I, sham operation group II, group III (model group), group IV [low dose concentrated beef broth group 200 mg / ( kg · d)], group V [medium dose concentrated beef broth 400 mg / (kg · d)] and group VI [high dose concentrated beef broth 600 mg / (kg · d)]. Group IV ~ group VI rats were fed with concentrated beef broth (200, 400 and 600 mg / (kg · d) respectively), and group II with 600 mg / (kg · d) concentrated beef broth. Groups I and Group III with the same amount of normal saline gavage. The neurological deficit scores of rats at 2 h and 7 d after operation were measured. Spleen mass index and volume of cerebral infarction were measured at 7 d after operation. The levels of TNF-α and IL-4 in the spleen were measured by enzyme linked immunosorbent assay (ELISA) Level. Results: Compared with group III, the scores of neurological deficit in group V and group VI decreased significantly at 2 h and 7 d after operation (P <0.05). At 7 d after operation, group I and group II rats Splenic index, spleen IL-4 and TNF-α expression was no difference; compared with group III, group V and group VI splenic index increased, the volume of cerebral infarction decreased, the level of TNF-α decreased, group VI rat spleen IL -4 expression was significantly increased (P <0.05). CONCLUSION: The concentrated beef broth can improve the neurological deficit, decrease the volume of cerebral infarction, raise the spleen index, decrease the level of TNF-α in the spleen and increase the spleen IL-4 in a dose-dependent manner.
