目的探讨不同浓度的胰升血糖素(Glg)和葡萄糖(Glu)对胰岛β细胞株MIN6中胰岛素(Ins)分泌功能及细胞内环磷鸟嘌呤核苷(cGMP)信号通路的影响。方法用不同浓度Glu(0、2.8、16.7 mmol/L)及Glg(0、100、500、1000 ng/L)处理MIN6细胞。ELISA测定细胞内cGMP含量,同时测定细胞上清液中Ins的含量。结果单纯Glg处理组中,1000 ng/L组cGMP含量较500 ng/L组、100 ng/L组低(P<0.05)。2.8 mmol/L Glu加不同浓度Glg处理组中,1000 ng/L Glg组cGMP含量较100 ng/L Glg组及0 ng/L Glg组低;500 ng/L Glg组较0 ng/L Glg组低(P<0.05)。16.7 mmol/L Glu加不同浓度Glg处理组中,1000 ng/L Glg组cGMP含量较其他组低;500 ng/L Glg组较100 ng/L Glg组及0 ng/L Glg组低(P<0.05)。单纯Glg处理组中,随Glg浓度升高,Ins分泌量升高(P<0.05)。2.8 mmol/L Glu加不同浓度Glg处理组中,1000 ng/L Glg组Ins较100 ng/L Glg组及0 ng/L Glg组高;500 ng/L Glg组Ins较0 ng/L Glg组高(P<0.05)。16.7 mmol/L Glu加不同浓度Glg处理组中,随Glg浓度升高,Ins分泌量升高(P<0.05)。结论 Glg可能通过浓度梯度的方式下调MIN6细胞内cGMP的产生从而促进MIN6细胞Ins分泌,且这种作用有Glu依赖性。
Objective To investigate the effects of different concentrations of Glg and Glu on insulin secretion and cGMP signal transduction in pancreatic β cell line MIN6. Methods MIN6 cells were treated with Glu (0,2.8,16.7 mmol / L) and Glg (0,100,500,1000 ng / L). The content of cGMP in the cells was measured by ELISA, and the content of Ins in the cell supernatant was also measured. Results Compared with 500 ng / L group and 100 ng / L group, the cGMP concentration of 1000 ng / L group was lower in Glg group (P <0.05). The concentration of cGMP in 1000 ng / L Glg group was lower than that in Gln group and 0 ng / L Glg group at 2.8 mmol / L Glu and Glg. Low (P <0.05). The concentration of cGMP in 1000 ng / L Glg group was lower than that in other groups at 16.7 mmol / L Glu and Glg treatment groups, but lower in 500 ng / L Glg group compared with 100 ng / L Glg group and 0 ng / L Glg group (P < 0.05). Glg alone treatment group, with the Glg concentration, Ins secretion increased (P <0.05). Insulin concentration in Insulin group was significantly higher than that in Gln group and Gln group at 100 ng / L and Gln group at 500 ng / L Glg High (P <0.05). In 16.7 mmol / L Glu plus different concentrations of Glg treatment group, with the Glg concentration increased, Ins secretion increased (P <0.05). Conclusion Glg may down-regulate the production of cGMP in MIN6 cells by concentration gradient and promote the secretion of Ins in MIN6 cells, and this effect is Glu-dependent.
