目的:本研究以目前国际病毒分类委员会(ICTV)所接受的肠道病毒分界标准为基础,通过客观地比较全基因组/多聚蛋白两两相似值,试图寻找一种简单可靠的方法来对新的肠道病毒进行的分类。方法:分析960株肠道病毒在全基因组水平上的两两比对相似值(pairwise identity scores,PIS)的频率分布图,观察在不同肠道病毒型别、不同种及不同属之间是否存在明显的分类界限,结合病毒聚类和系统发育树的结果寻找能够区分肠道病毒不同种/属的标准。结果:肠道病毒不同种间及与相近病毒属之间在全基因水平上存在比较好的PIS分类界限。多聚蛋白核苷酸PIS为0.657和0.536时可对肠道病毒进行可靠、稳定的种和属的分类。结论:肠道病毒多聚蛋白核苷酸序列间PIS在种和属水平上具备明显的遗传边界,并能够简单、可靠地用于肠道病毒监测实验室新种属的初步鉴定。“,”Objective:This study is based on the current enterovirus demarcation standards accepted by ICTV. By objectively comparing the whole genome/polyprotein pairwise identity score (PIS), an attempt was made to find a simple and reliable method to classify new enteroviruses.Methods:The distribution frequency of PIS in 960 strains of enteroviruses at the genome level was analyzed, and the existance of clear classification boundaries among different enterovirus types, different species and with other different genera were observed. The attempts were made to find the criteria that can distinguish different serotypes, species and genus of enteroviruses by combining the results of clustering and phylogenetic trees.Results:The frequency distribution of PIS showed that good PIS boundary between different enterovirus species and genus at the genome level existed. The PIS of polyprotein at 0.657 and 0.536 can be used for reliable and stable classification of enterovirus species and genera.Conclusions:The PIS of the nucleotide sequence of polyprotein showed obvious genetic boundary at the species and genus level, and can be used simply and reliably for the preliminary identification of new species and genus in enterovirus monitoring laboratory.
