目的同时测定蒺藜粗皂苷中海柯皂苷元、薯蓣皂苷元和替告皂苷元的量。方法采用HPLC-ELSD法,色谱柱为Agilent Zorbax SB-C18柱(250 mm×4.6 mm,5μm),流动相为甲醇-水(86∶14),漂移管温度100℃,载气体积流量2.5 L/min。结果海柯皂苷元、薯蓣皂苷元和替告皂苷元分别在114.1~1 141.0μg/mL(R2=0.999 2),16.88~168.80μg/mL(R2=0.999 4),78.60~786.00μg/mL(R2=0.999 3)呈良好线性关系;蒺藜粗皂苷中海柯皂苷元、薯蓣皂苷元和替告皂苷元的平均回收率分别为99.7%(RSD为1.6%)、99.3%(RSD为1.4%)、99.4%(RSD为1.1%)。结论该方法简便、准确、快速,且专属性良好,可用于蒺藜粗皂苷的质量评价。
Objective To determine the amount of hectorin, diosgenin and sapogenin in the crude extract of Tribulus terrestris. Methods HPLC-ELSD was used. The column was Agilent Zorbax SB-C18 column (250 mm × 4.6 mm, 5 μm). The mobile phase consisted of methanol-water (86:14), the drift tube temperature was 100 ℃, and the carrier gas volume flow was 2.5 L / min. Results The values of Hecogenin, diosgenin, and sapogenin were in the range of 114.1-114.0 μg / mL (R2 = 0.999 2), 16.88-168.80 μg / mL (R2 = 0.999 4), 78.60-786.00 μg / mL R2 = 0.999 3). The average recoveries of Hecogenin, Diosgenin and Sapogenin of Tribulus terrestris were 99.7% (RSD 1.6%), 99.3% (RSD 1.4%), 99.4% (RSD 1.1%). Conclusion The method is simple, accurate, rapid and specific, and can be used for the quality evaluation of the crude extract of Tribulus terrestris.
