目的研究乌头碱对体外培养的卵巢黄体细胞的毒性。方法无菌下制备25~29日龄雌性SD大鼠的卵巢黄体细胞,并通过透射电镜观察管嵴状线粒体来鉴定细胞。用MTT法检测0、0.05、0.5、5、50μg·mL-1的乌头碱对大鼠卵巢黄体细胞活力、激素分泌功能的影响,及对大鼠黄体细胞的氧化损伤作用。结果与溶剂对照组比较,作用24h后,50μg·mL-1乌头碱可明显抑制大鼠黄体细胞的增殖,差异有统计学意义(P<0.01);0.5、5、50μg·mL-1乌头碱对孕酮的分泌明显低于对照组(P<0.05)。结论乌头碱浓度高于5×102ng·mL-1时,对雌性大鼠卵巢黄体细胞有毒性作用,主要表现为抑制黄体细胞的增殖及抑制激素的分泌。
Objective To study the toxicity of aconitine on ovarian luteal cells cultured in vitro. Methods Ovarian luteal cells were prepared aseptically from 25 to 29 days old female Sprague-Dawley rats and the cells were identified by transmission electron microscopy of tube mitochondria. MTT assay was used to detect the effects of aconitine at 0, 0.05, 0.5, 5, and 50 μg·mL-1 on rat ovary luteal cell viability, hormone secretion, and oxidative damage to rat luteal cells. Results Compared with the solvent control group, 50 μg·mL-1 aconitine significantly inhibited the proliferation of rat luteal cells after 24 hours, and the difference was statistically significant (P<0.01); 0.5, 5, and 50 μg·mL-1. The secretion of progesterone from cephalospine was significantly lower than that of the control group (P<0.05). Conclusion When the concentration of aconitine is higher than 5×102 ng·mL-1, it has toxic effects on the luteal cells of female rats, and it mainly inhibits the proliferation of luteal cells and inhibits the secretion of hormones.
