目的 观察Obestatin腹腔注射对前壁心梗致慢性心衰(CHF)大鼠模型的心功能及其尿量的影响。方法 36只SD大鼠随机分为6组:对照组(n=6)及CHF模型组(n=30),后者包括:假手术组、精氨酸加压素(AVP)组、生长激素释放肽(Ghrelin)组、低剂量Obestatin组、高剂量Obestatin组(n均=6)。通过结扎心脏前降支建立CHF大鼠模型。16周后行心脏超声检查,并收集各组大鼠的24h尿量,随后每天1次腹腔注射:对照组注射生理盐水,其余5组大鼠分别注射生理盐水、AVP、Ghrelin以及低剂量Obestatin〔50 μg/(kg·d)〕和高剂量Obestatin〔100 μg/(kg·d)〕,注射4周后复查心脏超声并收集各组大鼠当天24 h尿量。结果 腹腔注射4周后,与注射前相比,高剂量Obestatin组大鼠心脏超声左心室射血分数(LVEF)显著升高,尿量显著增多(均P<0.05);AVP组大鼠LVEF显著下降(P<0.01),尿量显著下降(P<0.05);Ghrelin组大鼠心脏超声LVEF值显著升高(P<0.01),尿量无显著变化;对照组、生理盐水组、低剂量Obestatin组大鼠LVEF、尿量无显著变化。结论 高剂量Obestatin长期腹腔注射能明显改善心梗后心衰大鼠心功能,增加尿量。
Objective To observe the effects of Obestatin intraperitoneal injection on cardiac function and urine output of anterior wall myocardial infarction (CHF) rats. Methods Thirty - six SD rats were randomly divided into 6 groups: control group (n = 6) and CHF model group (n = 30). The latter included sham operation group, AVP group, Ghrelin group, low dose Obestatin group and high dose Obestatin group (n = 6). CHF rat model was established by ligating the anterior descending branch of the heart. Echocardiography was performed 16 weeks later. Urinary excretion of rats in each group was collected and then intraperitoneally injected once a day. Normal saline was injected into the control group, normal saline, AVP, Ghrelin and low dose Obestatin [50 μg (kg · d)] and high dose of Obestatin 〔100 μg / (kg · d)〕. After 4 weeks of injection, the echocardiography was performed and the urine output of the rats in each group was measured at 24 hours. Results After 4 weeks of intraperitoneal injection, the left ventricular ejection fraction (LVEF) and urine output in the high-dose Obestatin group were significantly increased (P <0.05 compared with those before injection). The LVEF in the AVP group was significantly higher than that before injection (P <0.01), and urine output decreased significantly (P <0.05). The LVEF of cardiac ultrasound in Ghrelin group was significantly increased (P <0.01), while the urine volume did not change significantly. The control group, saline group, low dose Obestatin There was no significant change in LVEF and urine volume in rats. Conclusion Long-term intraperitoneal injection of high-dose Obestatin can significantly improve cardiac function and increase urine output in CHF rats.
