目的:探讨胰高血糖素样肽-1(GLP-1)对小肠上皮细胞株IEC-6向胰岛素分泌细胞分化的作用。方法:采用不同浓度的GLP-1作用于IEC-6细胞24 h,MTT法检测细胞活性;GLP-1诱导IEC-6细胞5 d后,RT-PCR法检测胰腺十二指肠同源框1(PDX-1),胰岛素基因增强子结合蛋白1(Isl-1),神经源性分化蛋白(NeuroD)和胰岛素的mRNA表达,ELISA法检测培养基中是否存在胰岛素。结果:GLP-1可提高IEC-6细胞活性;使Isl-1和NeuroD的mRNA表达上调,并可以诱导PDX-1和胰岛素的表达;另外,GLP-1诱导5 d后培养基中可检测到胰岛素的表达。结论:GLP-1促进IEC-6细胞增殖,并可以诱导IEC-6细胞向胰岛素分泌细胞分化,而且PDX-1可能和GLP-1的促分化作用有关。
AIM: To investigate the effect of glucagon-like peptide-1 (GLP-1) on the differentiation of small intestine epithelial cell line IEC-6 into insulin-secreting cells. Methods: IEC-6 cells were treated with different concentrations of GLP-1 for 24 h, and the cell viability was measured by MTT assay. After IEC-6 cells were induced by GLP-1 for 5 days, the expression of GLP-1 in pancreatic duodenal homeobox 1 PDX-1, Isl-1, NeuroD and insulin mRNA were detected by enzyme-linked immunosorbent assay (ELISA). Results: GLP-1 increased the activity of IEC-6 cells. Up-regulated mRNA expression of Isl-1 and NeuroD and induced the expression of PDX-1 and insulin. In addition, GLP- Insulin expression. Conclusion: GLP-1 can promote the proliferation of IEC-6 cells and induce IEC-6 cells to differentiate into insulin-secreting cells, and PDX-1 may be related to the promoting differentiation of GLP-1.
