目的 筛查及分析胰腺导管腺癌中长链非编码RNA(lncRNA)及mRNA的差异表达谱,以及这些特异性lncRNAs的潜在通路、互作分子.方法 通过对5对胰腺导管腺癌/癌旁组织的芯片筛选及生物信息分析,构建共表达网络,结合国内外文献及多个数据库进行分析,预测lncRNA的潜在靶点、通路及互作关系.结果 按上调或下调5倍、P<0.01的标准,筛选得到癌及癌旁组织中差异表达的lncRNA 414个,mRNA 713个.迸一步生物信息学分析得到AC112229.1、RPL35AP、AC090377.1、UCA1、RP11-534L6.2、SAT1、AC027119.1、AK094786、BC042017、RP11-738E22.1、AC015818.5等11个可能参与胰腺导管腺癌发生发展关键环节的lncRNAs.结论 LncRNAs与胰腺导管腺癌的发生发展密切相关,深入相关研究对胰腺导管腺癌的诊断、治疗及预后判断具有重要意义.“,”Objective To analyze bioinformaticly the profile of surgical specimens pancreatic ductal adenocarcinoma (PDAC) to study the differential expressions of lncRNAs and mRNAs,and to predict the relative signal pathways and the biomolecular interactions of these specific lncRNAs.Methods The differential expressions of lncRNAs and mRNAs in specimens of PDAC were obtained using 5 pairs of microarrays.Then GO analysis,Pathway analysis and IncRNA-mRNA network were performed to predict the potential pathways and targets of these specific lncRNAs.The medical literatures and public databases on the internet were also searched.Results Using the standards that the expression of lncRNA/mRNA in PDAC to increase or reduce 5 times and a P value of <0.01,414 lncRNAs and 713 mRNAs were gained.Among them,11 lncRNAs (AC112229.1,RPL35AP,AC090377.1,UCA1,RP11-534L6.2,SAT1,AC027119.1,AK094786,BC042017,RP11-738E22.1,AC015818.5) were important in the development of PDAC on bioinformatic analysis.Conclusions LncRNAs are playing a significant role in PDAC.Further studies may show them to be important diagnostic,prognostic or therapeutic targets in the future.
