本文比较了氯胺-T法和Iodogen法碘化标记单克隆抗体BAC5在不同反应条件下的标记率,标记反应的时间与抗体免疫活性之间的关系。结果表明:这两种标记方法的最佳pH值为7~8;反应体积在100~200μl范围内均可以得到较好的结果;氯胺-T法反应3min,标记率已接近最大值,而Iodogen法要在10min后才有最大的产额。结果还表明:在本实验条件下,氨胺-T法和Iodogen法的反应时间分别小于3min和5min时,其抗体的免疫活性可保留原来的95%以上。
In this paper, chloramine-T method and Iodogen method labeled iodinated monoclonal antibody BAC5 under different reaction conditions labeling rate, labeling reaction time and antibody immunocompetence relationship. The results showed that the optimum pH values of these two methods were 7-8, and the reaction volume was within the range of 100-200μl. The chloramine-T method reacted for 3 minutes and the labeling rate was close to the maximum value. Iodogen law to have the largest output after 10min. The results also show that: under the experimental conditions, the reaction time of ammonia-T method and Iodogen method were less than 3min and 5min, the antibody immunological activity can retain more than 95% of the original.
