用大鼠及犬观察单次颈动脉灌注三尖杉酯碱的毒性反应。注药5mg/kg,大鼠有54%可耐受,46%于给药后12小时内死于循环呼吸衰竭。1.5和2mg/kg 致犬急性死于重要脏器出血,1mg/kg 犬可耐受,0.5mg/kg 为安全剂量。两种动物的主要毒性反应表现在给药时的窦性心动过速及心肌缺氧损害。致死动物的组织学检查可见多处心肌纤维断裂与脾脏白髓灶性坏死。存活大鼠观察4周,犬观察8周,其间未发现迟发的造血器官与肝、肾、心功能异常与神经系统症状。
Toxicity of harringtonine in single carotid artery perfusion of rats and dogs was observed. Injection of 5mg / kg, 54% of rats tolerated, 46% within 12 hours after administration died of circulatory failure. 1.5 and 2mg / kg to acute dogs died of major organ bleeding, 1mg / kg dogs can tolerate, 0.5mg / kg for the safe dose. The main toxicity of the two animals is manifested in the administration of sinus tachycardia and myocardial hypoxia damage. Histopathological examination of lethal animals revealed multiple myocardial fibrillation and focal necrosis of the white blood cells of the spleen. Survival rats observed for 4 weeks, dogs observed for 8 weeks, during which no delayed hematopoietic organs and liver, kidney, cardiac dysfunction and neurological symptoms were found.
