利用Northern印迹杂交技术对CCl4诱导大鼠肝纤维化过程中α1(Ⅰ)、α1(Ⅲ)及α1(Ⅳ)3种前胶原mRNA的表达状况进行观察。结果证实α1(Ⅰ)、α1(Ⅲ)及α1(Ⅳ)3种前胶原mRNA的表达均有增加,但三者的变化并不同步。纤维化早期首先是α1(Ⅳ)前胶原mRNA表达增加。随后是α1(Ⅲ)前胶原mRNA,后者在整个肝纤维化过程中呈优势性表达。α1(Ⅰ)前胶原mRNA的增加又滞后于α1(Ⅲ)且上升缓慢,至第20周时才逐渐与α1(Ⅲ)前胶原mRNA含量相近。可能是由于存在反馈性抑制的缘故,使得前胶原基因表达的上调并非呈直线递增而是曲线上升。
The expressions of α1 (Ⅰ), α1 (Ⅲ) and α1 (Ⅳ) procollagen mRNA in CCl4-induced hepatic fibrosis were observed by Northern blotting. The results confirmed that α1 (Ⅰ), α1 (Ⅲ) and α1 (Ⅳ) procollagen mRNA expression increased, but the three changes are not synchronized. Early fibrosis is the first α1 (Ⅳ) procollagen mRNA expression increased. Followed by α1 (Ⅲ) procollagen mRNA, which was predominantly expressed throughout the course of liver fibrosis. The increase of α1 (Ⅰ) procollagen mRNA lags behind that of α1 (Ⅲ) and slowly increases, and gradually increases to the level of α1 (Ⅲ) procollagen mRNA at the 20th week. Possibly due to the presence of feedback inhibition, the upregulation of procollagen gene expression did not increase linearly but rather curve up.
