目的:探讨柔肝通络汤对大脑中动脉阻断(MACO)所致脑缺血/再灌注损伤大鼠脑细胞凋亡及神经元形态的影响。方法:将实验大鼠分为柔肝通络汤高、中、低剂量组和空白对照组、假手术组、模型组。采用线栓法制备大鼠局灶性脑缺血/再灌注损伤模型,采用TUNEL法及Nissl染色法分别观察实验动物缺血再灌注后3、6、12、24、48h脑凋亡细胞及神经元形态。结果:柔肝通络汤能够显著降低模型动物的凋亡细胞数,增加活神经元数量。柔肝通络汤组的凋亡细胞数低于模型组(P<0.05),活神经元数量高于模型组(P<0.05)。结论:柔肝通络汤通过抗细胞凋亡发挥脑保护作用。
Objective: To investigate the effects of Rougan Tongluo Decoction on cerebral cell apoptosis and neuron morphology in rats with cerebral ischemia / reperfusion injury induced by middle cerebral artery occlusion (MACO). Methods: The experimental rats were divided into Rougan Tongluo Decoction high, medium and low dose groups and blank control group, sham operation group and model group. The model of focal cerebral ischemia / reperfusion injury was established by thread occlusion method. TUNEL and Nissl staining were used to observe the apoptotic cells and nerves at 3, 6, 12, 24 and 48 hours after ischemia / reperfusion Metamorphosis Results: Rougan Tongluo Decoction can significantly reduce the number of apoptotic cells in model animals and increase the number of living neurons. The number of apoptotic cells in Rougan Tongluo Decoction group was lower than that in model group (P <0.05), and the number of viable neurons was higher than that in model group (P <0.05). Conclusion: Rougan Tongluo Decoction can exert cerebral protective effect through anti-apoptosis.
