目的研究氯化锰对大鼠生精细胞细胞色素C(cytochrome-c,cyto-c)和支持细胞波形蛋白(vimentin,VM)表达的影响及对生精功能的抑制效应。方法将雄性SD大鼠随机分为空白对照、低剂量和高剂量组,8只/组。MnCl_2组分别染锰4和6周,空白对照组给予等容生理盐水,给药途径均为腹腔注射,取睾丸采用免疫组织化学(SABC)法检测生精细胞cyto-c和支持细胞VM表达,测定睾丸脏器系数,取附睾检测精子数量和精子畸形率。结果与空白对照组比较,各染锰组生精细胞cyto-c阳性率由80%下降至60%,VM阳性率由50%下降至2%,睾丸脏器系数由1.2%下降至1%,精子数量由1.1×10~7/ml下降至0.2×10~7/ml,精子畸形率由28‰上升至64‰,且均呈一定的时间-效应关系和剂量-效应关系。各组大鼠精子数量与cyto-c和VM阳性率均呈正相关。结论锰可诱导大鼠cyto-c和VM表达,抑制生精功能。
Objective To investigate the effect of manganese chloride on the expression of cytochrome-c (cyto-c) and vimentin (VM) in rat spermatogenic cells and its inhibitory effect on the function of spermatogenesis. Methods Male SD rats were randomly divided into blank control, low dose and high dose group, 8 / group. The MnCl_2 group was incubated with Mn for 4 and 6 weeks, respectively, and the blank control group was given isocratical saline. The route of administration was intraperitoneal injection. The testis was used to detect the cyto-c and VM expression in spermatogenic cells by immunohistochemistry (SABC) Determination of testicular organ coefficient, take sperm test sperm count and sperm deformity rate. Results Compared with the blank control group, the positive rate of cyto-c in spermatogenic cells decreased from 80% to 60%, the positive rate of VM decreased from 50% to 2%, the testicular organ coefficient decreased from 1.2% to 1% The number of sperm decreased from 1.1 × 10-7 / ml to 0.2 × 10-7 / ml, and the sperm deformity rate increased from 28 ‰ to 64 ‰, showing a time-effect relationship and dose-effect relationship. The number of sperm in each group was positively correlated with the positive rate of cyto-c and VM. Conclusion Manganese can induce cyto-c and VM expression in rats and inhibit the function of spermatogenesis.
