目的:采用HPLC法测定不同配比黄连干姜水煎液中盐酸小檗碱含量的变化。方法:色谱柱为安捷伦C18硅胶柱(150mm×4.6mm,5μm),流动相为乙腈-0.01mol·mL-1磷酸二氢铵水溶液-磷酸(20∶79.9∶0.1),流速为1mL·min-1,检测波长为345nm,进样量为20μL。结果:盐酸小檗碱在10~200μg/mL范围内与峰面积呈良好的线性关系(r=0.999 9),平均回收率为99.93%,方法精密度RSD为1.19%,各杂质峰与主峰分离度良好。结论:不同配比黄连干姜中盐酸小檗碱的含量均高于黄连单煎液,且黄连干姜比例为6∶1时水煎液中盐酸小檗碱的含量最高。
Objective: To determine the content of berberine hydrochloride in different proportion of Coptis chinensis decoction by HPLC. METHODS: The column was an Agilent C18 silica gel column (150 mm × 4.6 mm, 5 μm) with a mobile phase of acetonitrile-0.01 mol · mL -1 ammonium phosphate monobasic phosphate (20:79.9: 0.1) and a flow rate of 1 mL · min- 1, the detection wavelength of 345nm, injection volume of 20μL. Results: Berberine hydrochloride showed a good linear relationship with the peak area (r = 0.999 9) in the range of 10 ~ 200 μg / mL with the average recovery of 99.93%. The RSD of the method was 1.19% Degree is good. Conclusion: The contents of berberine hydrochloride in Coptis chinensis with different proportions were all higher than those with Coptis chinensis alone, and the content of berberine hydrochloride in decoction was the highest when the ratio of Coptis chinensis was 6:1.
