从1例终末期骨髓瘤患者的胸水中,用浆细胞特异性单抗-B-B_4免疫磁珠筛选的方法分离出骨髓瘤细胞,纯度大于99%,连续体外培养2个月建株。所用细胞传代培养基中含5ng/ml的IL_6和5ng/ml的TNF。该株细胞被命名为OH-2。OH-2细胞EB病毒阴性,分泌单克隆IgGκ链,表达PCA-1、CD_(38)、B-B_4、CD_(56)和两种TNF受体(P~(55)、P~(75)),而B系的早期细胞分化抗原如CD_(10)、CD_(19)、CD_(20),仅在有些细胞表面有微弱表达。经DNA指纹图谱分析证实,OH-2细胞系是来自患者骨髓癌细胞。
Myeloma cells were isolated from the pleural effusion of one patient with end-stage myeloma using myeloid-specific monoclonal antibody B-B_4. The purity of myeloma cells was more than 99%. Cell subculture media used contained 5 ng / ml of IL-6 and 5 ng / ml of TNF. The strain was named OH-2. OH-2 cells were negative for Epstein-Barr virus (EBV) and secreted monoclonal IgG kappa chain, which expressed PCA-1, CD_38, B_4, CD_5 and two TNF receptors (P55, ), While the early differentiation of B cells such as CD_ (10), CD_ (19) and CD_ (20) were only weakly expressed on some cell surface. DNA fingerprinting analysis confirmed that the OH-2 cell line was derived from the patient’s bone marrow cancer cells.
