目的:研究结肠癌患者外周血基质金属蛋白酶11(MMP-11)、癌胚抗原(CEA)、细胞角蛋白20(CK-20)、CD44mRNA的表达特点及意义。方法:采用RT-PCR检测外周血及组织样本各肿瘤标志物基因m RNA表达,其中初诊结肠癌患者、晚期结肠癌患者各20例外周血样本10mL(试验组),上述20例晚期结肠癌患者手术切除的癌组织样本(阳性对照组),健康体检者20例外周血样本10mL(阴性对照组),分析其表达的差异。结果:结肠癌患者外周血中MMP-11、CEA、CK-20、CD44mRNA水平明显高于健康体检者(P<0.05);晚期结肠癌患者癌组织中各肿瘤标志物水平明显高于其外周血中的四种肿瘤标志物水平(P<0.05);结肠癌DukesC-D期患者外周血中各肿瘤标志物水平明显高于DukesA-B期患者(P<0.05);低分化结肠癌患者外周血中各肿瘤标志物明显高于高、中分化者(P<0.05)。结论:外周血MMP-11、CEA、CK-20、CD44mRNA表达水平可为结肠癌的早期诊断、筛查、分期提供有意义的参考。
Objective: To investigate the expression and significance of matrix metalloproteinase 11 (MMP-11), carcinoembryonic antigen (CEA), cytokeratin 20 (CK-20) and CD44mRNA in peripheral blood of patients with colon cancer. Methods: RT-PCR was used to detect the mRNA expression of tumor markers in peripheral blood and tissue samples, including 10 mL of 20 peripheral blood samples from newly diagnosed colon cancer patients and advanced colon cancer patients (experimental group). The above 20 patients with advanced colon cancer Surgical resection of cancer tissue samples (positive control group), healthy subjects 20 cases of peripheral blood samples 10mL (negative control group), analysis of the differences in expression. Results: The levels of MMP-11, CEA, CK-20 and CD44 in peripheral blood of patients with colon cancer were significantly higher than those of healthy controls (P <0.05). The levels of tumor markers in patients with advanced colon cancer were significantly higher than those in peripheral blood (P <0.05). The levels of tumor markers in peripheral blood of patients with DukesC-D colon cancer were significantly higher than those of DukesA-B patients (P <0.05). Conclusion: The expression of MMP-11, CEA, CK-20 and CD44mRNA in peripheral blood can provide a meaningful reference for the early diagnosis, screening and staging of colon cancer.
