目的:优选北虫草可溶性蛋白质的最佳提取工艺。方法:对北虫草蛋白质提取条件进行单因素考察,并采用L9(34)正交试验设计法及考马斯亮蓝染色法,以北虫草可溶性蛋白质含量为指标,考察p H值、提取温度、料液比、提取时间4个因素对可溶性蛋白质提取率的影响,从而确定北虫草蛋白质的最佳提取工艺。结果:建立了北虫草蛋白质的最佳提取工艺:向北虫草药材粉末中加入60倍量p H为10的蒸馏水,在温度30℃条件下回流提取70 min。结论:优化的提取工艺稳定、合理、可行,能正确反映北虫草药材中的蛋白质含量,为北虫草生物活性物质的进一步研究提供了实验依据。
Objective: To optimize the best extraction process of soluble protein of Cordyceps militaris. Methods: The single factor study on the extraction conditions of Cordyceps militaris protein was carried out. The L9 (34) orthogonal design method and Coomassie brilliant blue staining method were used to evaluate the soluble protein content of Cordyceps militaris. The effects of p H value, extraction temperature, Ratio, the extraction time of four factors on the extraction rate of soluble protein to determine the best Cordyceps protein extraction process. Results: The optimal extraction process of the protein of Cordyceps militaris was established. Sixty times the amount of distilled water with pH value of 10 was added to the powder of Cordyceps militaris and the mixture was refluxed for 70 min at 30 ℃. Conclusion: The optimized extraction process is stable, reasonable and feasible, which can correctly reflect the protein content of Cordyceps militaris. It provides an experimental basis for the further study of the bioactive substance of Cordyceps militaris.
