本文采用丝素作为酶的固定化材料 ,以丝网印刷一次性碳电极为基础电极 ,经化学修饰二茂铁后 ,将含有多酚氧化酶的蘑菇萃取蛋白固定于其表面以制作酶电极。该酶电极具有良好的分析性能 ,对其底物邻苯二酚或多巴胺有良好的响应 ,线性范围为 2× 10 -8mol/L至 2× 10 -2 mol/L ,响应速率快 ,在 30秒内即可达到95 %稳态电流 ,该电极有较好的重现性 ,变异系数为 2 .7% ,且可保持 10天以上的工作寿命。该电极在富含水分的低温环境中可保存较长的时间 ,因此可望在此研究基础上实现其商品化
In this paper, silk fibroin was used as the immobilized material of the enzyme. Screen-printed disposable carbon electrode was used as the basic electrode. After the chemical modification of ferrocene, the mushroom extract containing polyphenol oxidase was immobilized on the surface to make enzyme electrode. The enzyme electrode has good analytical performance, good response to its substrate catechol or dopamine, the linear range of 2 × 10 -8mol / L to 2 × 10 -2 mol / L, the response rate is fast, at 30 95% of the steady state current can be achieved in seconds, the electrode has good reproducibility, the coefficient of variation of 2.7%, and can maintain the working life of more than 10 days. The electrode can be stored for longer periods of time in a moisture-rich, low-temperature environment and is therefore expected to commercialize on the basis of this study
