本研究利用Cre/LoxP系统构建B细胞条件性敲除Senp1基因小鼠模型(CKO),探讨了SUMO特异性蛋白酶1(SENP1)对C57BL/6小鼠B细胞数量以及对血清免疫球蛋白产生的作用。利用流式细胞术检测对照组(WT)及实验组(CKO)小鼠骨髓、脾脏及淋巴结B细胞含量及分布情况,结果显示,Senp1敲除后可显著降低脾脏和淋巴结中B细胞的比例,而对骨髓中的B细胞比例没有影响。利用ELISA方法检测血清中免疫球蛋白IgM、IgG、IgG3及IgG1亚型的水平,结果显示CKO小鼠血清IgG1的含量明显低于WT小鼠,而IgM、IgG及IgG3含量没有显著差别。以上结果提示SENP1在小鼠外周淋巴器官B细胞分布及IgG1产生过程中可能起调控作用。
In this study, B cell conditional knockout of Senp1 mouse model (CKO) was constructed by using Cre / LoxP system. The effects of SUMO specific protease 1 (SENP1) on the number of B cells and serum immunoglobulin production in C57BL / 6 mice effect. The contents and distribution of B cells in bone marrow, spleen and lymph node of control group (WT) and experimental group (CKO) were detected by flow cytometry. The results showed that the proportion of B cells in spleen and lymph node was significantly decreased after Senp1 knockdown, While the proportion of B cells in the bone marrow has no effect. Serum immunoglobulin IgM, IgG, IgG3 and IgG1 subtypes were detected by ELISA. The results showed that the serum IgG1 level in CKO mice was significantly lower than that in WT mice, while the levels of IgM, IgG and IgG3 were not significantly different. The above results suggest that SENP1 may play a regulatory role in the distribution of B cells in peripheral lymphoid organs and the production of IgG1 in mice.
