酒女贞子抗氧化谱效关系研究

目的对酒女贞子特征图谱与抗氧化活性的相关性进行研究,确定酒女贞子抗氧化活性物质基础。方法运用HPLC测定经典恒温加速试验处理的酒女贞子的特征图谱,并通过1,1-二苯基-2-三硝基苯肼(DPPH)、2,2’-联氨-双(3-乙基苯并噻唑啉-6-磺酸)二胺盐(ABTS)和FRAP法测定其抗氧化活性,运用偏最小二乘回归分析法进行数据分析,探究酒女贞子量化特征图谱和抗氧化活性的相关性,筛选活性化合物相关色谱峰。结果从17个匹配特征图谱中筛选出峰1、峰3、峰8、峰10、峰14和峰15与酒女贞子清除DPPH自由基、ABTS自由基能力呈显著正相关;峰3、峰8、峰12和峰13与酒女贞子还原Fe~(3+)能力显著正相关。其中峰3和峰8与酒女贞子清除DPPH自由基、ABTS自由基和还原Fe~(3+)能力均正相关。并确定了峰3为红景天苷、峰12为木犀草苷、峰13为特女贞苷、峰14为橄榄苦苷、峰15为女贞苷、峰17为木犀草素。结论酒女贞子药效不是由特女贞苷和红景天苷等个别指标成分的含量而决定。全面分析与药效相关的化合物群组的含量才能真实反映酒女贞子的质量。 Objective To study the correlation between the characteristic map of Fructus Aurantii and antioxidative activity and to determine the substance basis of antioxidant activity of Fructus Aurantii. Methods The characteristic chromatogram of Ligustrum lucidum treated by classical thermostatic accelerated test was determined by HPLC and analyzed by HPLC with DPPH and 2,2’-hydrazine-bis (3 -benzothiazoline-6-sulfonic acid) diamine salt (ABTS) and FRAP method were used to analyze the antioxidant activity. The data were analyzed by partial least-squares regression analysis to explore the quantitative characteristic map and anti- Oxidative activity of the relevant active compounds screening related peaks. Results The peak 1, peak 3, peak 8, peak 10, peak 14 and peak 15 were screened from 17 matched profiles. There was a significant positive correlation between ABPH free radical and DPPH free radical scavenging capacity. Peak 3, peak 8, peak 12 and peak 13 with the Ligustrum lucidum restore Fe ~ (3 +) capacity was significantly positively correlated. Peak 3 and peak 8 were positively correlated with the ability of Fructus Lycii to scavenge DPPH, ABTS and Fe (3 +). And identified peak 3 salidroside, peak 12 is luteolin, peak 13 is a special rosynin, peak 14 is oleuropein, peak 15 is the rosette, peak 17 is luteolin. Conclusion The effect of Fructus Ligustri Lucidi is not determined by the content of individual index components such as virginia and salidroside. A comprehensive analysis of the efficacy of the compound group content to truly reflect the quality of the wine Ligustrum.
