目的应用SNPs的复合PCR扩增体系检验实际案件中遇到的含脱落细胞检材、腐败检材及骨组织等疑难检材,为疑难检材的检验提供一种新的检验方法。方法通过用IdentifilerTM试剂盒及已建立的47个SNPs(包括1个性别位点)位点复合扩增荧光检测体系,并行对照检验36起案件中的人体脱落细胞、陈旧血痕、高度腐败的肌肉、软骨以及牙齿、骨等各类疑难检材共109份,对于人体脱落细胞、陈旧血痕、牙齿、骨等检材,STR与SNPs的检验成功率均达到80%以上,SNPs略高于STR。对于高度腐败的肌肉及软骨检材,SNPs的检验成功率明显高于STR。结论所建立的SNPs复合PCR扩增体系在疑难检材检验中具有较高的实用价值。
Objective To detect complex exfoliated cell samples, corrosive samples and bone tissues encountered in real cases by using SNPs multiplex PCR amplification system to provide a new test method for the test of difficult samples. Methods The fluorescent detection system was amplified by using IdentifilerTM kit and 47 established SNPs (including one sex locus). The control samples were tested in 36 cases of human exfoliated cells, old bloodstains, highly corrupted muscles, Cartilage and dental, bone and other kinds of difficult to detect a total of 109 samples for human exfoliated cells, old bloodstains, teeth, bone and other samples, STR and SNPs test success rate reached more than 80%, SNPs slightly higher than the STR. For highly corrupted muscle and cartilage samples, the success rate of SNPs testing was significantly higher than that of STR. Conclusion The established SNPs multiplex PCR amplification system has high practical value in the test of difficult samples.
