有机氚化物对离体培养小鼠胎儿中脑细胞增殖及分化的影响

作者就四种有机氚化物对体外培养细胞的增殖及分化的影响进行了研究。方法细胞取自11日胎龄小鼠中脑组织,对其进行高密度细胞培养,并加入有机氚化物与培养液。结果中脑细胞的分化比增殖对辐射更敏感。辐射引起剂量依赖性的培养细胞DNA和蛋白质含量的减少。以各指标50%的抑制剂量,用X射线为参考辐射估算的氚RBE值在4.6~8.7范围。混合辐射,特别是当使用3H-TdR时,能更有效地抑制中脑细胞的分化。0.05Gy即可产生可检测到的抑制效应。结论高密度中脑细胞培养系统对辐射极为敏感,来自有机氚化物的β射线对该系统中细胞的增殖及分化的抑制作用与参考辐射X射线相比较,具有极高的相对生物效应比值。 The authors studied the effects of four organic tritides on the proliferation and differentiation of cultured cells in vitro. Methods The cells were collected from the midbrain of 11-day-old mice and cultured in high-density cells with organic tritium and culture medium. Results The differentiation of midbrain cells is more radiation sensitive than proliferation. Radiation induced a dose-dependent reduction of cellular DNA and protein content. The RBE value of tritium estimated with X-ray as reference radiation ranged from 4.6 to 8.7 with 50% inhibition of each indicator. Mixed radiation, especially when using 3H-TdR, can more effectively inhibit mesencephalic cell differentiation. 0.05Gy can produce a detectable inhibitory effect. Conclusions High density mesobrain culture system is extremely sensitive to radiation. The β ray from organic tritium has a very high relative bioavailability compared with the reference radiation X-ray in inhibiting the proliferation and differentiation of cells in this system.
